摘 要:建立一种采用微生物显色法对动物源性食品中抗生素残留进行初筛,之后再结合高效液相色谱-质谱(high performance liquid chromatography-mass spectrometry,HPLC-MS)法对阳性样品进行复测的分析检验方法.利用大肠杆菌(E.coli)作为指示菌,制备96微孔板,选取pH 7.4磷酸钠-乙腈缓冲液进行提取,以反应液150 μL、菌液50 μL(初始A600 nm0.4左右)、样品提取液100 μL作为检测体系,可同时检测15 种喹诺酮类药物.结果表明:所建立的微生物显色法操作简便、快捷、成本低,结果准确且易判断,对动物源性食品中喹诺酮类药物检出限为40~200 μg/kg,符合国内外对抗生素残留限量的要求.对50 份动物源性样品进行检测,其结果与HPLC-MS复测结果一致,说明该微生物法稳定性良好,可用于动物源性食品中喹诺酮类药物残留的初筛检测.

Abstract: A new microbial chromogenic assay was developed to analyze antibiotic residues in animal-derived food and after that, the positive samples were identified by a high-performance liquid chromatography-tandem mass spectrometric (HPLC-MS) method. E.coli was used as the indicator strain to produce a 96-well microplate for detecting 15 kinds of quinolones. Muscle samples were extracted by pH 7.4 phosphate-acetonitrile buffer. This system of microbial chromogenic assay consisted of 150 μL the reaction solution, 50 μL of bacterial suspension (the initial A600 nm value was around 0.4), and

100 μL of the extract solution. The assay proved to be simple and cheap and the results were accurate and easy to interpret. The detection limits for 15 kinds of quinolones drugs were 40?200 μg/kg, meeting the domestic and international requirements for the determination of antibiotic residue limits. Consistent results were obtained for 50 samples of animal-derived food by microbial chromogenic assay and HPLC-MS, indicating that the assay could be used to detect antibiotic residues in animal-derived food with good stability.

喹诺酮类药物(quinolones,QNs)是人工合成的具有1,4-二氢-4-氧代喹啉-3-羧酸结构的一类药物总称[1].该药作为抗生素使用已有超過50 年的历史,主要通过阻断特异性拓扑酶,抑制细菌DNA复制从而发挥抗菌作用[2],该药具有价格低廉、抗菌谱广、无交叉耐药性等优点,广泛应用在临床及养殖领域.但喹诺酮类药物常有不合理使用和滥用的情况发生,使其或其代谢产物残留于动物的肌肉和脏器组织中,进而通过食物链导致人体产生抗药性,影响疾病治疗与康复,引起严重的食品安全问题和出口贸易问题[3].因此,喹诺酮类药物残留问题越来越引起人们的重视.
