摘 要:建立了同时测定食用菌中3种荧光增白剂(C.I.24,C.I. 210,C.I. 220)含量的高效液相色谱法.样品前处理采用50% N,N-二甲酰胺-水溶液作提取剂,常温超声提取30 min.采用Sepax BR- C18柱为分析柱,以10 mmol·L-1甲酸铵水溶液和甲醇为流动相进行梯度洗脱,荧光激发波长为350 nm,发射波长为435 nm.结果显示,3种荧光增白剂在1~200 ng·mL-1浓度范围内相关系数均大于0.999,线性范围较宽,线性关系良好;C.I.24检出限为15 μg·kg-1,定量限为30.0 μg·kg-1,C.I.210和C.I.220检出限为7.5 μg·kg-1,定量限为15.0 μg·kg-1;3种荧光增白剂不同添加水平的加标平均回收率在75.1%~108.1%,RSD在1.2%~14.4%.该方法操作简便,结果准确,灵敏度高,重现性好,能够适用于食用菌中荧光增白剂的检测.

Study on Determination Method of Fluorescent Whitening Agents in Edible Fungi by HPLC

Abstract: A method for the determination of three kinds of fluorescent whitening agents(C.I.24, C.I. 210, C.I. 220)in edible fungi by HPLC was developed. The samples were extracted with 50% N, N-Dimethyl formamide-water solution by ultrasonic for 30 min at room temperature. The HPLC method was performed on a Sepax BR- C18 column, and 10 mmol·L-1 Ammonium formate and methanol were used as mobile phases in the gradient elution. Fluorescence detector was adopted in the excitation welength of 350 nm and emission welength of 435 nm. The results showed that the correlation coefficient of three kinds of fluorescent whitening agents in the range of concentration of 1 ~ 200 ng·mL-1 were greater than 0.999. The limit of detection of C.I.24 was 15 μg·kg-1, and the limit of quantification was 30.0 μg·kg-1, the limit of detection of C.I.210 and C.I.210 were 7.5 μg·kg-1, and the limit of quantification were 15.0 μg·kg-1. The erage recoveries of three kinds of fluorescent whitening agents were in the range of 75.1%~108.1% at different spiked levels, and the RSD were in the range of 1.2%~14.4%. This method was simple, accurate, high sensitive and reproducible, and could be applied to the determination of fluorescent whitening agents in edible fungi.

熒光增白剂是一种荧光染料,它通过吸收不可见紫外光,再发射出可见蓝光或蓝紫色荧光,可使被染物品达到增白增艳的效果,被广泛应用于纺织、纸张、洗涤剂、包装等制造加工业[1].除增白作用外,荧光增白剂还具有一定的防腐保鲜功能,因此市场上有不法商家将其添加到双孢菇、海鲜菇等白色食用菌中,以使产品色亮好看,货架期延长.另外,在包装、运输和销售过程中,食用菌与包装材料接触也可能导致荧光增白剂污染.有报道称,荧光增白剂被人体吸收后经长期积累会有致癌风险[2].虽然现在荧光增白剂对人体的危害问题尚无定论,但基于目前的试验和研究水平,有必要对荧光增白剂的使用进行限制和监管.

在我国,荧光增白剂被划分为精细化工产品,严禁违法添加到农产品中.国家卫生标准也已明确规定,食品包装用原纸、餐具洗涤剂、食品工具设备用洗涤剂与洗涤消毒剂中均不做到检出荧光性物质.为保障消费者的健康安全,在加强对食用菌中荧光增白剂污染问题监管力度的同时,研究和制定食用菌中荧光增白剂含量的测定方法有着重要的现实意义.目前,有关食用菌中荧光增白剂检测方法并无相关国家标准,现行的农业行业标准NY/T 1257—2006只能作定性检测,不能作定量检测,在一定程度上限制了食用菌安全监管效果.有关荧光增白剂检测方法的研究也主要集中在食品包装材料[3]、纸张[4]、洗涤剂[5]及塑料制品[6]等方面,关于食用菌的报道较少.有文献报道[7],采用离子对试剂进行液相检测,能有效分离检测3种荧光增白剂,但离子对试剂的使用会造成色谱柱不可逆的损伤,缩短其使用寿命;另有报道[8],采用液质联用法,结果准确、灵敏度高,但该法质谱设备成本高、操作技术要求高、固相萃取前处理步骤繁琐,方法推广有一定的局限性.
